Rao was born on  30 June 1939 to Kanakam and Seshamma in Bhimavaram, West Godavari District, Andhra Pradesh, India. He is wed to R. Sashiprabha, a former King George Hospital administrator. Neurosurgeon Sunkara Balaparameswara Rao is his brother.

He was a student at U.L.C.M. High. From Andhra Medical College in Visakhapatnam, he earned his M.B.B.S. in 1966 and his Master of Orthopaedic Surgery in 1970.
